Marker genes

AREG, CD83, CSRNP1, DNAJB1, DNAJB9, DUSP2, NR4A1, NR4A2, PLAUR, PPP1R15A, RASGEF1B, RNASE6, SELENOK, SGK1, SLC2A3, TACSTD2, ZFP36, ZNF331

Annotation genes

CD83, DUSP2, NR4A1, NR4A2, ZFP36

Annotation rationales

CD83 marks terminal maturation of cDC1 and efficient T-cell–interacting immunological synapses. DUSP2 tempers MAPK signaling, while NR4A1/NR4A2 provide transcriptional “brakes” that prevent hyperactivation after PRR/TNF signals. ZFP36 accelerates decay of inflammatory mRNAs (e.g., TNF/IL1), fine-tuning output during antigen encounter; co-expression of stress chaperones (DNAJB1/9) and PLAUR implies membrane and cytoskeletal remodeling needed for migration and cross-talk.

Functional implications

Executes a burst of antigen processing and cross-presentation with controlled cytokine release; migrates to T-cell areas to prime CD8⁺ responses while using DUSP2/NR4A1/NR4A2/ZFP36 to avoid immunopathology. Supports productive licensing of CTLs, coordinated chemokine secretion, and tissue egress toward lymph nodes.
