Annotation rationales
Chronic low-grade self or environmental BCR engagement induces NR4A1/2/3, TNFAIP3 (A20), and NFKBID (IκBNS)—a tolerance/attenuation circuit that dampens NF-κB/AP-1 while preserving viability. CD83 marks “poised” APC interface but absence of GC/plasmablast modules confirms hyporesponsiveness. BCL2A1/SELENOK/SQSTM1 support survival and antioxidant/autophagy buffering to maintain this restrained state.
Functional implications
Enforces self-tolerance in the naïve compartment: maintains hyporesponsiveness to weak/self antigens, reduces bystander activation in inflamed tissues, yet keeps cells alive and relocatable when proper costimulation appears. This protects against autoimmunity while preserving a recruitable pool for bona fide primary responses when context (Tfh help, PAMPs) validates activation.
