Annotation genes
ATF3, CD69, DUSP1, FOS, JUN
Annotation rationales
CD69 upregulation with FOS/JUN/EGR1 (IEGs) and ATF3 denotes acute BCR and/or cytokine stimulation. DUSP1 provides MAPK negative feedback to prevent over-shoot, while DNAJB1/HSPA1A/B indicate a chaperone surge that stabilizes receptors and nascent secretory proteins under proteotoxic load. GADD45B/DDIT3 reflect stress-adapted survival/transcriptional rewiring; RHOB supports endosomal trafficking/immune synapse remodeling. Altogether this captures naïve B cells within minutes–hours of priming, before fate bifurcation.
Functional implications
Acts as the early activation gate for primary responses: rapidly enhances antigen capture and costimulation, withstands folding stress as synthesis ramps, and tunes BCR thresholds via phosphatase feedback. Functionally, this subset triages cells toward either (i) GC entry for affinity maturation or (ii) extrafollicular plasmablast differentiation for immediate low-affinity antibody—while retaining the option to revert to quiescence if help is insufficient.
