Marker genes

CHST2, COTL1, CXCR4, HLA-DPB1, HLA-DQA1, HLA-DQB1, HLA-DRA, HLA-DRB1, HLA-DRB5, MYBL2, RFTN1

Annotation genes

CHST2, CXCR4, HLA-DRA, HLA-DRB1, MYBL2

Annotation rationales

MYBL2 drives proliferation, while CXCR4 mediates retention in CXCL12-rich stromal zones. High MHC-II expression (HLA-DRA, HLA-DRB1) is unusual in plasma cells and indicates a transitional plasmablast stage capable of antigen presentation. CHST2 enhances lymphoid homing via selectin ligand synthesis. This profile captures a proliferative, migratory intermediate between B cells and long-lived plasma cells.

Functional implications

At this transitional stage, cells proliferate to expand the IgA-secreting pool while simultaneously presenting antigen to T cells, reinforcing cognate B–T collaboration. Their CXCR4 expression ensures retention in niches where survival factors such as IL-6 and APRIL are abundant. Functionally, they bridge adaptive responses and terminal differentiation, seeding long-lived IgA plasma cells while providing short-term antigen presentation capacity within mucosa-draining lymphoid tissues.
