Marker genes
CALCR, CDH15, CXCR4, ITGA7, MYF5, NCAM1, PAX7, SPRY1, VCAM1
Annotation genes
CXCR4, ITGA7, PAX7, SPRY1, VCAM1
Annotation rationales
Classic satellite-cell niche identity anchored by PAX7 (stemness TF). Adhesion receptors ITGA7 (laminin α2 in basal lamina), VCAM1, NCAM1, and CDH15 maintain tight apposition to myofibers. CXCR4 retains cells near CXCL12⁺ vessels/fibroblasts, optimizing oxygen and trophic support. SPRY1 is a MAPK antagonist that enforces quiescence and preserves long-term self-renewal, while CALCR (calcitonin receptor) adds an anti-activation brake. Low MYF5 flags early myogenic priming without full commitment.
Functional implications
Sustain a quiescent-but-ready stem-cell reservoir tightly attached to laminin-rich niches. Sense chemokine/morphogen gradients (CXCR4/SPRY1) to decide between dormancy and activation, enabling rapid, synchronized recruitment into the repair program when damage occurs.
