Annotation genes

RPL11, RPS6, TPT1

Annotation rationales

A translationally upshifted, cell-cycle-permissive state: broad induction of ribosomal subunits (e.g., RPL11, RPS6) and the growth/anti-apoptotic factor TPT1 indicates ramped protein synthesis to support proliferation, cytoskeletal remodeling, and secretory output. This profile commonly appears as microglia clonally expand near lesions or adapt to metabolic stress; RPS6 also reflects mTOR pathway engagement that fuels process outgrowth and phagolysosome biogenesis during activation.

Functional implications

Rapid pool expansion and phenotypic switching: locally proliferate to repopulate niches after neuronal/glial injury, scale up proteostasis and lysosomal components for forthcoming debris clearance, and prepare for state transitions (e.g., to phagocytic/APC microglia). Contribute to scar remodeling and restoration of homeostatic microglial density while minimizing excitotoxicity by timely resolution of the activated program.
