Marker genes
CALCA, CLDN7, CRIP2, EDNRB, EEF1A2, EPCAM, FOXA2, HSPB8, LNCpress1, OLIG1, PLD1, POU5F1, PPIC, RARRES2, SLC7A11, TFF3, TRH, VWA5A
Annotation genes
CLDN7, EPCAM, FOXA2, TFF3
Annotation rationales
The epithelial/endoderm triad EPCAM, CLDN7, and lineage TF FOXA2 together with secretory goblet-associated TFF3 marks a definitive endoderm–leaning epithelium emerging from pluripotency (still retaining POU5F1 in markers). Such profiles are typical after low-dose Activin A/RA exposure in vitro. Inclusion of CLDN7, EPCAM, FOXA2, TFF3 emphasizes epithelialized endoderm priming.
Functional implications
Serves as an efficient source for gut tube/hepato-pancreatic progenitors under Activin/Nodal → WNT/FGF induction; rapidly matures into foregut/midgut-like epithelium with mucus/secretory competence, improving yields of hepatic, biliary, and intestinal lineages.
