Annotation genes
CCR7, CD83, CXCR4, MIR155HG, MYC
Annotation rationales
CD83 marks recent activation and enhanced APC interactions; BCL2A1 secures survival during the transition to proliferation. MYC/MIR155HG program rapid cell-cycle entry and effector polarization, consistent with imminent plasmablast generation or GC re-entry. CCR7↔CXCR4 provides niche-hopping between T-cell zones, follicles, and bone marrow, enabling staged access to Tfh help, antigen, and survival factors. NR4A family calibrates activation thresholds to prevent overshoot, and NFKBID (IκBNS) shapes BCR-dependent gene expression during recall. This state typifies memory cells “on the launchpad.”
Functional implications
Acts as a rapid-deployment recall force that can very quickly expand, form stable synapses with Tfh, and either seed secondary germinal centers for renewed affinity maturation or differentiate into high-output plasmablasts. Chemokine flexibility allows precise routing to the microanatomical locale most favorable for the next step—T cell–rich zones for licensing, follicle for selection, or marrow for long-term residence. By combining survival insurance (BCL2A1) with growth drivers (MYC/miR-155), these cells deliver high-magnitude secondary antibody responses while preserving a portion of the pool for sustained memory.
