Annotation rationales

MHC-II molecules (HLA-DRA/DRB1, DP, DQ) with CD74 are striking in CD8 T cells, reflecting a state of aberrant or induced antigen-presentation capacity, often triggered by strong cytokine exposure (IFNγ, type-I IFN). APOBEC3G suggests antiviral restriction activity; CD27 supports a memory-like background. In parallel, GZMA/GZMK reveal acquisition of cytotoxic function, indicating these cells sit at the interface of antigen presentation and killing. This mixed program suggests transitional cells capable of signaling to CD4 T cells while exerting effector functions, potentially amplifying immune responses in inflamed tissue.

Functional implications

Operates as a hybrid APC–cytotoxic population: presents peptide–MHC II to CD4 T cells while delivering granzyme-mediated cytotoxicity. Functionally, these cells may enhance local T cell–T cell communication, prolong antigen availability, and expand CD4 helper responses during infection. At tissue entry sites or inflamed niches, they serve as amplifiers of adaptive immunity, boosting both effector CD8 activity and Tfh/CD4 help, at the cost of higher activation stress and potential autoimmune risk.
