Marker genes
CCL3, CCL4, CCL4L2, DNAJB1, GADD45B, GZMK, HSPA1B, IER3, IFNG, RASD1, TAGAP, TNF
Annotation genes
CCL4, GZMK, IFNG, TNF
Annotation rationales
Chemokines CCL3/4 and cytokines TNF and IFNG mark a locally activated, inflammation-amplifying Trm state. GZMK adds cytotoxic potential with a slower, pro-inflammatory bias compared to GZMB. DNAJB1/HSPA1B chaperones and GADD45B/IER3 stress-response genes indicate adaptation to inflammatory tissue niches. TAGAP/RASD1 contribute to TCR and cytokine signaling fine-tuning. This profile captures Trm engaged in active orchestration of tissue inflammation.
Functional implications
Functions as a signal-amplifying Trm subset that rapidly produces inflammatory cytokines and chemokines to recruit myeloid and lymphoid partners (monocytes, NK, T cells). By broadcasting danger signals, they transform tissue niches into immune activation hot spots, enhancing pathogen clearance but also carrying risk of bystander tissue damage if unrestrained.
