Marker genes

ANXA1, EZR, IL7R, LMNA, MYADM, RGCC

Annotation genes

EZR, IL7R, RGCC

Annotation rationales

IL7R expression ensures access to IL-7–mediated survival cues, a central feature of long-lived Trm. EZR and RGCC support cytoskeletal remodeling and cell-cycle restraint, compatible with tissue anchoring and quiescence. ANXA1, LMNA, MYADM stabilize plasma membrane and nuclear integrity, aligning with a maintenance phenotype rather than active effector function. Together, these markers define a quiescent Trm pool that persists in tissues awaiting reactivation.

Functional implications

Provides the homeostatic backbone of Trm populations, ensuring persistence for years at barrier sites. These cells maintain tissue residency through adhesion and survival pathways, consuming minimal resources while retaining readiness. On challenge, they can rapidly re-enter effector programs or proliferate locally, preserving tissue-protective immunity long after infection or vaccination.
