Annotation rationales
A classic repair/“de-differentiated” Schwann-cell program: ATF3, EGR1, IER3, and NR4A1 are immediate-early injury regulators that drive exit from a myelinating state and entry into a pro-regenerative phenotype; CCL2 recruits macrophages/monocytes to clear myelin debris; CRYAB (αB-crystallin) provides chaperone-mediated protection during proteotoxic and oxidative stress; MAFF and CEBPD integrate redox/cytokine cues; RCAN1 modulates calcineurin–NFAT signaling to restrain excessive inflammation. This constellation mirrors the well-described inflammatory-reactive Schwann state that orchestrates Wallerian degeneration and sets the stage for axonal regrowth.
Functional implications
Rapidly transitions myelinating fibers into a debris-clearing, growth-permissive niche: secretes CCL2 and other cues to recruit/educate macrophages, upregulates chaperones (CRYAB) to protect damaged proteins, and remodels ECM to form Bands of Büngner that guide regenerating axons. Temporarily suppresses myelin gene expression to prioritize clearance and protection, then hands off to remyelinating programs once axons re-extend. By calibrating calcineurin/NR4A1-linked signaling, limits bystander damage and restores immune–glial homeostasis after peripheral nerve injury or inflammatory neuropathy.
