Marker genes

APP, CD9, CNP, GPRC5B, LAMP2, MAL, NDRG2, OPALIN, PLLP, QDPR, RNASE1, SPOCK1, SUN2

Annotation genes

CNP, MAL, OPALIN, PLLP

Annotation rationales

CNP, MAL, OPALIN, and PLLP are core components of the mature myelin program. CNP supports cytoplasmic channels and microtubule–actin dynamics in the inner tongue; MAL and PLLP promote compact myelin and lipid raft organization; OPALIN is highly enriched in myelinating OLs and links to paranodal/myelin membrane specialization. Together with vesicle/trafficking (CD9, LAMP2) and axon-interaction molecules (APP), this profile reflects fully differentiated oligodendrocytes actively building and stabilizing CNS myelin.

Functional implications

Generate thick, electrically insulating sheaths with precise node/paranode organization, increasing conduction velocity and fidelity across long-range cortical and subcortical tracts. Continuous vesicle delivery sustains sheath growth and turnover, while metabolic coupling (e.g., lactate shuttling) supports axonal health and spike reliability.
