Annotation rationales
As neural-crest derivatives, melanocytes retain neurotrophic guidance logic. NTRK2 (TrkB) and NRP2 (semaphorin/VEGF co-receptor) promote dendrite elongation and directional motility in response to neurotrophins/semaphorins within the epidermal–follicular niche. FRZB (sFRP3) tempers WNT tone, a key rheostat for switching between migration and pigment production. GPX3 provides extracellular antioxidant protection against UV-generated peroxides bathing the melanocyte–keratinocyte unit. Lipid transporter ABCA8 suggests membrane remodeling during neurite-like extension.
Functional implications
Extend long dendrites across keratinocyte fields to optimize melanin distribution; dynamically tune WNT levels via FRZB to balance migration vs melanin output; integrate neurotrophin/semaphorin cues through NTRK2/NRP2 to refine placement around follicles and rete ridges; buffer extracellular ROS with GPX3 to protect both melanocytes and neighboring keratinocytes during sun exposure.
