Annotation genes
CEBPA, CLEC12A, CTSG, FCGR1A, MPO
Annotation rationales
CEBPA is the lineage-defining TF for granulocytic commitment; MPO (myeloperoxidase) and CTSG (cathepsin G) are classic azurophilic granule enzymes of neutrophil precursors; FCGR1A (CD64) and CLEC12A (MICL) denote maturing myeloid immunoreceptors typical of GMPs. Concurrent granule biogenesis and receptor acquisition indicates cells poised for neutrophil differentiation while retaining monocyte potential.
Functional implications
Generate neutrophils and monocytes by driving CEBPA-dependent myelopoiesis, building primary granules (MPO/CTSG), and installing high-affinity Fc receptors (FCGR1A) and inhibitory checkpoints (CLEC12A) for calibrated innate responses. Rapidly expand frontline antimicrobial capacity.
