Marker genes
BIK, CD24, CHAD, CLDN11, DEGS2, GJA4, GRP, GSTM3, IL20RA, ISYNA1, ODC1, SBSPON, SCG3
Annotation genes
CLDN11, GJA4
Annotation rationales
CLDN11 is a tight-junction protein specific to LEC valve leaflets; GJA4 (Cx37) is required for valve morphogenesis and mechanoelectrical coupling. CHAD/SBSPON contribute to anchorage of leaflet matrix; BIK/ODC1/ISYNA1 reflect controlled turnover and membrane synthesis in valve cells subjected to oscillatory shear. Together this is the canonical valve endothelium program.
Functional implications
Unidirectional flow enforcers: maintain leaflet cohesion and sealing, synchronize opening/closure via Cx37-mediated coupling, and adapt junctional composition to oscillatory shear. They prevent backflow, minimize energy loss in lymph propulsion, and protect downstream nodes from pressure spikes during muscle pumping or respiratory cycles.
