Marker genes
ACTB, ADIRF, CCL21, FSCN1, IGF1, IGFBP2, IGFBP5, IL32, RHOC
Annotation genes
CCL21, FSCN1, IGF1
Annotation rationales
CCL21 is a hallmark chemokine of initial/tip LECs guiding CCR7⁺ DCs/T cells; FSCN1 (actin bundling) and RHOC/ACTB drive filopodia/lamellipodia for sprouting and tissue probing. IGF1/IGFBPs add trophic support and survival under low shear. This constellation maps to migratory/tip-like LECs at expanding fronts of the lymphatic capillary bed.
Functional implications
Pathfinders and immune guides that extend toward chemotactic gradients, shepherd CCR7⁺ leukocytes to afferent routes, and initiate new lymphatic sprouts during edema resolution or tissue remodeling. They lay permissive tracks in ECM while preserving CCL21 highways for efficient antigen delivery to draining nodes.
