Marker genes

BTG1, CDKN1A, DNAJB1, FOS, GADD45B, HES1, HSPA1A, JUN, JUNB, NFKBIA, PPP1R15A, RGS16, RHOB, SOCS3, ZFP36

Annotation genes

DNAJB1, FOS, HSPA1A, JUN, NFKBIA, ZFP36

Annotation rationales

IEG/AP-1 (FOS/JUN/JUNB) and NF-κB brake (NFKBIA) indicate minutes–hours acute activation; ZFP36/PPP1R15A/SOCS3 impose anti-inflammatory/translational brakes to prevent runaway signaling. HSPA1A/DNAJB1 mark a chaperone surge that protects junctional and valve proteins during shear/cytokine stress. CDKN1A/BTG1 transiently restrain the cell cycle, a hallmark of endothelial alarm states. Together this is a classic inflammatory gate-opening LEC profile tuned to remain reversible and protective.

Functional implications

Rapid-response LECs that dilate initial lymphatics, upshift antigen and leukocyte trafficking, and transiently loosen cortical actin to increase cell transit while deploying chaperones to preserve barrier and valve leaflet integrity. They integrate chemokine cues with inhibitory feedback (ZFP36/SOCS3) to resolve inflammation and re-establish low-leak, low-pressure lymph flow once danger subsides.
