Marker genes
CBLB, CD36, GDF15, MAML2, MAPRE2, P4HA1, P4HA3, PHLDA1, UBC
Annotation genes
CD36, GDF15, P4HA1
Annotation rationales
CD36 drives fatty acid uptake and lipid accumulation; GDF15 is a stress-induced cytokine elevated in lipotoxicity, signaling metabolic distress to hepatocytes/systemic circuits. P4HA1/3 support collagen hydroxylation, enabling new ECM deposition. This cluster reflects LSECs adapting to lipid overload with fibrogenic remodeling.
Functional implications
Lipotoxicity-activated remodelers: increase fatty acid uptake and act as stress messengers (via GDF15), while reinforcing sinusoidal ECM with new collagen. This contributes to capillarization and fibrosis in NAFLD/NASH, impairing filtration and exacerbating metabolic dysfunction.
