Marker genes
ARHGAP6, BASP1, BDKRB1, CCBE1, HAS1, PTGES, SEMA3C, TREM1
Annotation genes
BDKRB1, CCBE1, HAS1, PTGES, SEMA3C, TREM1
Annotation rationales
PTGES drives PGE synthesis (pro-inflammatory/pro-algesic), HAS1 increases hyaluronan production altering pericellular viscosity and cell–matrix signaling, and BDKRB1 (bradykinin receptor) links to pain and vascular permeability. TREM1 amplifies myeloid-type inflammatory cascades, while SEMA3C/CCBE1 provide neuro/lymphangiogenic cues. Together this reflects chondrocytes sending angiogenic and neurotrophic signals atypical for immune-privileged cartilage.
Functional implications
Pro-angiogenic, neurotrophic chondrocytes that foster vessel and nerve ingrowth at osteochondral junctions, particularly in the degenerating knee. Consequences include increased pain sensitization, PGE-driven synovitis, altered HA-rich matrix hydration, and potential breach of the avascular cartilage niche. Functionally, they promote osteochondral crosstalk and accelerate structural progression of OA by enabling vascular/immune access to cartilage.
