Annotation rationales

Adipocytes exposed to cytokines, lipolytic hormones, or fasting mount a stress/inflammatory program. ANGPTL4 promotes lipolysis and modulates vascular/immune interactions; SOCS3 dampens JAK–STAT signaling (often elevated by IL-6), contributing to insulin resistance. IFITM3, MAFF, and MYC indicate interferon/redox/activation responses; NAMPT (visfatin/NAD salvage) supports metabolic stress adaptation and can signal to immune cells. EMP1 and S100A10 mark activation and membrane dynamics seen in cytokine-responsive adipocytes. Classic lipid-storage markers are relatively low, consistent with an activated, catabolic state.

Functional implications

Coordinate immuno-metabolic defense in the depot: increase lipolysis and free-fatty-acid release (ANGPTL4), secrete inflammatory mediators/alarmins, and impose negative feedback on cytokine signaling (SOCS3). Shape macrophage/T cell recruitment and promote depot-wide insulin resistance during stress or obesity, while sustaining NAD metabolism (NAMPT) for survival.
