Annotation rationales
Signature lacks terminal adipocyte markers (e.g., CIDEA/PLIN1/ADIPOQ) and is enriched for trafficking/proteostasis (STX12, PSMD8, CDC37L1), RNA processing (MBNL2), and metabolic priming (PGM3). NRIP1 (RIP140) represses mitochondrial/oxidative programs and UCP1, typical of cells prior to full maturation. MYLIP (IDOL) regulates LDLR abundance, tuning cholesterol flux in differentiating adipocytes. Together this fits preadipocyte/early committed adipocyte poised to differentiate rather than a fully lipid-loaded cell.
Functional implications
Provide a progenitor/transition pool that can proliferate modestly and differentiate upon PPARγ/insulin cues into mature white or beige adipocytes. Functions include membrane/vesicle remodeling and ECM interaction necessary for adipogenesis; endocrine activity is limited, with metabolic programs held in check by NRIP1 until differentiation proceeds.
